Cannot publish Asp.Net Core application on IIS

I'm trying to publish my app on IIS. It gives the below error while clicking on Test Settings.

There was an error while performing this operation. Details: Invalid application path.

Steps:

1. Open IIS.
2. Right-click Default Web Site and select Add Application.
3. Give Alias and select DefaultAppPool as application pool and select the physical path MyProject\bin\Release\PublishOutput.
4. Click on Test Settings.

But If I try the below steps, I'm successfully able to publish my app.

1. Open IIS.
2. Right-click Sites and then select Add Web Site.
3. Give Alias and select DefaultAppPool as application pool and select the physical path MyProject\bin\Release\PublishOutput.
4. Click on Test Settings.

Please let me know if I'm doing anything wrong.

IIS 6, Asp.Net Core 2.0.1, Visual Studio 2017 version 15.6.6

Update:

I tried to enable logs in the application. the log file is generated when I try to browse the application. It says Application startup exception: MySql.Data.MySqlClient.MySqlException (0x80004005): Unknown database 'spardb'. But the spardb database exists. So what could be the possible root cause?
